KPV (Lys-Pro-Val) is a C-terminal tripeptide fragment of alpha-melanocyte-stimulating hormone (α-MSH) that retains potent anti-inflammatory activity. This research peptide modulates inflammatory signaling through NF-κB pathway suppression and melanocortin receptor engagement. KPV is studied in preclinical models of gut inflammation, immune cell activation, and inflammatory cytokine regulation.
KPV represents the minimal anti-inflammatory signaling sequence derived from the 13-amino acid α-MSH peptide. Research has established that the C-terminal tripeptide (residues 11-13) retains the anti-inflammatory activity of the full-length hormone without significant melanogenic or melanocortin receptor-mediated metabolic effects. KPV enters cells and inhibits NF-κB signaling by preventing nuclear translocation of the p65 subunit, reducing transcription of pro-inflammatory genes including TNF-α, IL-1β, IL-6, and COX-2. Published research has demonstrated KPV’s anti-inflammatory efficacy in colitis models, demonstrating reduced mucosal inflammation and improved epithelial barrier function (Dalmasso et al., PLoS ONE, 2008). KPV inhibits NF-κB activation by interfering with IκB kinase (IKK) activity, preventing IκB phosphorylation and subsequent p65 nuclear translocation. This reduces expression of NF-κB-dependent pro-inflammatory genes. Additionally, KPV modulates MAPK signaling cascades, reducing p38 and JNK phosphorylation in activated immune cells. The peptide’s anti-inflammatory effects have been demonstrated to be both melanocortin receptor-dependent and receptor-independent, with direct intracellular action on NF-κB machinery.
KPV provides targeted NF-κB anti-inflammatory activity from the melanocortin system, while BPC-157 acts through angiogenesis and NO signaling.
