PT-141 (Bremelanotide) is a synthetic cyclic heptapeptide melanocortin receptor agonist derived from the α-MSH analog Melanotan II.
PT-141 was developed from Melanotan II through metabolic conversion and subsequent direct synthesis. While Melanotan II acts broadly across melanocortin receptor subtypes (MC1R-MC5R), PT-141 demonstrates preferential activity at MC3R and MC4R, the subtypes most relevant to central nervous system signaling related to sexual behavior. The peptide activates MC4R in the hypothalamic paraventricular nucleus and medial preoptic area, triggering downstream dopaminergic signaling through the mesolimbic pathway. PT-141 preferentially targets MC3R/MC4R for central neurobehavioral effects, while MT-2 (Melanotan II) activates all five melanocortin receptor subtypes. MT-1 (Afamelanotide) is selective for MC1R (pigmentation). Oxytocin acts through its own receptor system rather than melanocortin pathways.
